چکیده

Mathematical models are presented which attempt to relate the interplay between such factors as life span of a leukemic animal, how early drug treatment is initiated, the survival of sensitive and resistant leukemic cells in treated animals, the incidence of mutation from the sensitive state to the resistant state, and the dihydrofolate reductase levels in mixed populations of sensitive and resistant cells. A computer was used to generate theoretical mixed populations of resistant and sensitive leukemic cells during three simulated passages in drug-treated animals. This was done so as to mimic the actual enzyme data collected during the development of an amethopterin-resistant subline. It is concluded that dihydrofoÃ-atereducÃ-asecan be used as a meaningful bio chemical marker in studies of emerging antifolate-resistant leukemic cells.

Quantitation of dihydrofolate reductase in individual parental and methotrexate-resistant murine cells. Use of a fluorescence activated cell sorter.

A fluorescent labeling technique has been devised to quantitate dihydrofolate reductase levels in individual cells and has been used to examine the heterogeneity in dihydrofolate reductase levels within various populations of methotrexate-resistant and parental cells. Precise quantitation of cellular dihydrofolate reductase levels in parental and methotrexate-resistant cells (having up to sever...

Genetic mutations in 57 and 58 codons gene of Plasmodium vivax dihydrofolate reductase

Introduction: The use of Sulfadoxine and pyrimethamine (SP) for treatment of vivax malaria is not common in most of malarious areas because of sensivity of this parasite to chloroquine. But, Plasmodium vivax isolates are exposed to SP because of mixed infection with P.falciparum and this subject has lead to emergence of mutations in P.vdhfr gene. As Plasmodium vivax is the most prevalent specie...

Relative frequency and kinetic properties of transport-defective phenotypes among methotrexate-resistant L1210 clonal cell lines derived in vivo.

Information was sought on the relative extent to which transport-defective, methotrexate-resistant phenotypes emerge among the total subpopulation of resistant phenotypes during therapeutic challenge of leukemic cells in vivo. A number of monoclonal methotrexate-resistant sublines of the L1210 leukemia were derived during methotrexate therapy of leukemic mice and biochemically characterized. Of...

Selective Multiplication of Dihydrofolate Reductase Genes in Methotrexate-resistant Variants of Cultured Murine Cells*

The rate of dihydrofolate reductase synthesis in the AT3000 line of methotrexate-resistant murine Sarcoma 180 cells is approximately 200to 250-fold greater than that of the sensitive, parental line. We have purified cDNA sequences complementary to dihydrofolate reductase mRNA and subsequently used this probe to quantitate dihydrofolate reductase mRNA and gene copies in each of these lines. Anal...

Treatment of leukemia with large doses of methotrexate and folinic acid: clinical-biochemical correlates.

Patients with acute leukemia were given repeated cycles consisting of infusions of methotrexate followed by "rescue" with folinic acid. Peripheral blood leukemic cells were harvested from patients before cyclical treatment, and the rates of incorporation of thymidine and of deoxyuridine into deoxyribonucleic acid (DNA) were measuared in vitro. There was no relationship between the pretreatment ...
